Genome-wide inventory and functional characterization of cannabinoid oxidocyclases in Cannabis sativa
Horticulture Research
Surveys five cannabis genomes for cannabinoid oxidocyclases, the enzymes that convert CBGA into THCA, CBDA and CBCA, and identifies 36 putatively functional genes in five clades, including a divergent gene the authors name THCAS-like 1 (TASL1). Expression in mature glandular trichomes was profiled across nine varieties and correlated with cannabinoid content across 21. Assays in Pichia pastoris and Nicotiana benthamiana showed TASL1 and CBDAS-like 1 producing mainly THCA and CBDAS-like 2 varying with the host, and substitutions in an oxygen-reactivity motif raised the catalytic efficiency of CBDAS up to threefold. The paper is a short research letter.
